TEPEZZA®
Company: Amgen
Therapeutic Area: Immunology, Rare Diseases
Drug Type: Biologic
Molecule: Teprotumumab-trbw
Clinical Phase: Approved
Pipeline Status: Active
Indication: Thyroid Eye Disease (TED), also known as Graves’ Orbitopathy. It is indicated for both active and chronic TED regardless of disease duration.
Mechanism of Action: Mechanism of Action (MoA) TEPEZZA is a first-in-class Insulin-like Growth Factor-1 Receptor (IGF-1R) Inhibitor. 1. Targeting the Root: In patients with TED, IGF-1R is overexpressed on orbital fibroblasts. 2. Action: By binding to and blocking IGF-1R, TEPEZZA inhibits the signaling complex that drives inflammation, fat expansion, and muscle fibrosis behind the eye. 3. Results: This leads to a significant reduction in proptosis (eye bulging), diplopia (double vision), and orbital pain/swelling.
