Lilly Indiana Manufacturing Investment of $4.5 Billion, Opens Dedicated Genetic Medicine Facility

Lilly Indiana manufacturing investment is expanding again, with Eli Lilly, and Company announcing an additional $4.5 billion commitment across two of its three manufacturing sites in Lebanon, Indiana. The announcement brings Lilly’s total capital expansion commitments in Indiana since 2020 to more than $21 billion. The company said the additional investment is being driven by its evolving pipeline and anticipated demand for its medicines.

The investment covers Lilly Lebanon API, described by the company as one of its future active pharmaceutical ingredient sites, and Lilly Lebanon Advanced Therapies, which opened May 6 as Lilly’s first dedicated genetic medicine manufacturing facility. The new facility is designed to support both clinical and commercial production of advanced therapies targeting disease at the genetic level.

Lilly said the advanced therapies facility will cover a full spectrum of genetic medicine modalities, extending from research-stage development through large-scale commercial supply. Building the facility required the development of new manufacturing processes because the modalities involved did not have established commercial manufacturing precedent.

The Lebanon campus is planned to contain three sites: Lilly Lebanon Advanced Therapies, Lilly Lebanon API and the Lilly Medicine Foundry. The company describes Lebanon, Indiana, as the cornerstone of its domestic manufacturing buildout. Lilly previously announced plans for Zepbound (tirzepatide) and Mounjaro (tirzepatide) production at the Lebanon API site, and the latest investment expands the manufacturing commitment to additional medicines.

Lilly Indiana Manufacturing Investment Expands Advanced Medicine Production

The latest Lilly Indiana manufacturing investment includes planned production of Foundayo (orforglipron), which Lilly identifies in the release as its FDA-approved once-daily oral treatment for weight loss, as well as retatrutide, an investigational triple hormone receptor agonist being studied in late-stage development for obesity and cardiometabolic disease.

The manufacturing expansion therefore connects Lilly’s Indiana infrastructure with multiple areas of its medicine portfolio. The company says its Lebanon API site is expected to support production of medicines including Zepbound, Mounjaro, Foundayo and retatrutide.

According to Lilly Chair and CEO David A. Ricks, the company’s goal is not only to discover medicines but also to build advanced manufacturing plants capable of producing them. Lilly said the Lebanon API site is expected to open in 2027 and would become the largest API production site in U.S. history.

The company’s broader U.S. capital expansion commitments since 2020 now total more than $50 billion, according to the release. Lilly also said it plans to break ground on several recently announced U.S. manufacturing sites during 2026.

Lilly Lebanon Advanced Therapies Opens for Genetic Medicine Manufacturing

Lilly Lebanon Advanced Therapies represents a new manufacturing capability within Lilly’s Indiana network. The facility is intended to support advanced therapies that target disease at the genetic level and is designed for both clinical production and commercial-scale manufacturing.

The company said the facility incorporates manufacturing processes developed specifically for genetic medicine modalities for which there was no established commercial precedent. Its intended scope extends from research-stage development through large-scale commercial supply, positioning the site as part of Lilly’s effort to build manufacturing capacity alongside its evolving pipeline.

The facility is the first of three planned sites on the Lebanon campus. Lilly Lebanon API and the Lilly Medicine Foundry are planned as the other two components of the campus.

Zepbound, Mounjaro and Foundayo Manufacturing Plans

Lilly’s Lebanon manufacturing expansion includes several medicines highlighted in the company’s release. Zepbound and Mounjaro, both based on tirzepatide, were previously identified by Lilly for production at the Lebanon API site.

The company describes Zepbound as a dual GIP and GLP-1 receptor agonist approved for certain adults with obesity or overweight with weight-related medical problems, and also for adults with moderate-to-severe obstructive sleep apnea and obesity.

Mounjaro is described as an injectable medicine containing tirzepatide for adults and children 10 years and older with type 2 diabetes, used with diet and exercise to improve blood glucose.

The new investment also includes planned production of Foundayo (orforglipron). Lilly describes Foundayo as a once-daily, non-peptide oral GLP-1 receptor agonist for chronic weight management. The company states that it can be taken without restrictions on food or water intake.

Lilly also identifies retatrutide as part of the manufacturing expansion. Retatrutide remains investigational and is being evaluated in Phase 3 clinical trials across obesity, type 2 diabetes and other conditions listed by the company.

Economic Impact of Lilly’s Indiana Expansion

The Lilly Indiana manufacturing investment also extends beyond the company’s manufacturing sites, according to the release. Lilly cited findings from an Indiana University Kelley School of Business, Indiana Business Research Center report concerning Lilly’s economic and civic contributions in Indiana.

The company said the report found that Lilly accounts for 70% of Indiana’s pharmaceutical GDP and that every Lilly job supports more than two additional jobs across the state. Lilly also estimates that each dollar it spends in the area can generate up to four dollars in additional local economic activity.

Indiana officials and Indiana University leadership cited the investment as contributing to the state’s life sciences and advanced manufacturing sectors, while Lilly characterized its Indiana expansion as part of a broader domestic manufacturing strategy.
